SFPQ
SFPQ (Splicing factor, proline- and glutamine-rich) is a protein that switches other genes on and off. The public catalogues list it as a fusion partner and a DNA repair gene, and the evidence so far is association rather than a proven role.
Overview
DNA- and RNA binding protein, involved in several nuclear processes. Essential pre-mRNA splicing factor required early in spliceosome formation and for splicing catalytic step II, probably as a heteromer with NONO. Binds to pre-mRNA in spliceosome C complex, and specifically binds to intronic polypyrimidine tracts.
Open Targets scores its association with cancer at 0.57 (direct and indirect evidence; datatypes literature 0.90, genetic association 0.00, somatic mutation 0.89).

- 3 · How drugs use it
No product in this corpus aims at SFPQ yet. Transcription factors have no pocket to plug, so drugs either degrade them or block the partner protein they need to dock on DNA.

Biology
DNA- and RNA binding protein, involved in several nuclear processes. Essential pre-mRNA splicing factor required early in spliceosome formation and for splicing catalytic step II, probably as a heteromer with NONO. Binds to pre-mRNA in spliceosome C complex, and specifically binds to intronic polypyrimidine tracts. Involved in regulation of signal-induced alternative splicing. During splicing of PTPRC/CD45, a phosphorylated form is sequestered by THRAP3 from the pre-mRNA in resting T-cells; T-cell activation and subsequent reduced phosphorylation is proposed to lead to release from THRAP3 allowing binding to pre-mRNA splicing regulatotry elements which represses exon inclusion. Interacts with U5 snRNA, probably by binding to a purine-rich sequence located on the 3' side of U5 snRNA stem 1b. Location: Nucleus speckle; Nucleus matrix; Cytoplasm (UniProt). Locus 1p34.3 (HGNC).
Notes
top- Written by scripts/fetch-cancer-genes.ts from CIViC, Open Targets, IntOGen, HGNC and UniProt; the function text is UniProt's, condensed and in UK spelling. Roles: UniProt disease notes describe a translocation or gene fusion involving the gene; UniProt keyword "DNA repair". Evidence tier "association-only" is the strongest of those signals.
- Prevalence not recorded: none of the sources gives a positivity rate.
