ABC-07
ABC-07 is the UK trial asking whether focused high-dose radiotherapy after chemotherapy helps people whose bile duct or gallbladder cancer has not spread but cannot be removed. Twelve UK centres took part and its final results are awaited.
Overview
ABC-07 (ISRCTN10639376) is a randomised phase 2 in the UK ABC series for locally advanced biliary tract cancer. Patients receive six cycles of cisplatin and gemcitabine followed by stereotactic body radiotherapy (up to 50 Gy in 5 fractions or 67.5 Gy in 15 fractions) or eight cycles of cisplatin and gemcitabine; the primary outcome, updated in August 2026, is progression-free survival from randomisation to progression on CT or MRI by RECIST 1.1 or death. The target was 83 patients, the registry end date is 12 August 2026, and the radiotherapy quality assurance paper (Clinical Oncology 2026) reports that 12 UK centres recruited and 41 patients were randomised to the radiotherapy arm, with protocol deviations in 46 percent of plans, mostly unavoidable target coverage compromise because of the duodenum and other nearby organs. Sites listed include the Churchill Hospital Oxford, University College Hospital, the Royal Marsden, Mount Vernon, the Lister Hospital, the Royal Free, Velindre, St Bartholomew's, Hammersmith, Southampton General and Addenbrooke's. For gallbladder cancer with hilar involvement it is the UK counterpart of the Indian RUGB trial.
