CARTITUDE-6
CARTITUDE-6 asks whether a single CAR-T infusion can replace the stem cell transplant that has anchored myeloma treatment for thirty years, when both follow the same four-drug induction.
Overview
CARTITUDE-6 (also EMN28) is a phase 3 trial of 759 transplant-eligible patients with newly diagnosed multiple myeloma. All receive DVRd induction; one arm then proceeds to high-dose melphalan and autologous stem cell transplant with DVRd consolidation, the other to a single infusion of the BCMA-directed CAR-T ciltacabtagene autoleucel, and both go on to lenalidomide maintenance. The primary endpoints are progression-free survival and sustained minimal residual disease negativity.
The trial is running and has not reported. Its answer will decide whether transplant remains the standard consolidation for fit patients or gives way to cellular therapy.
