DRAMMATIC asks two questions about maintenance treatment after a stem cell transplant for myeloma: whether adding daratumumab to lenalidomide helps people live longer, and whether those whose tests show no detectable disease after two years can safely stop; it is still running with about 1,100 patients.
Overview
DRAMMATIC (SWOG S1803) is a US National Clinical Trials Network phase 3 trial in patients with multiple myeloma after autologous stem cell transplant, randomised to lenalidomide maintenance or lenalidomide with subcutaneous daratumumab. After two years, patients who are MRD-negative are randomised a second time to continue or stop maintenance, making the trial one of the first to test MRD-guided discontinuation. The primary endpoint is overall survival, assessed for up to 15 years.
The registry lists an estimated 1,100 participants, active and no longer recruiting, with primary completion in 2032 and no results. The corpus's multiple myeloma page cites DRAMMATIC with MASTER for MRD-guided discontinuation in trials.
