Blacksmith Medicines designs drugs against metal-containing enzymes. In cancer it is developing inhibitors of FEN1, a DNA-repair enzyme, to combine with PARP inhibitors in ovarian cancer, and the work is in the laboratory.
Blacksmith Medicines has a chemistry platform for medicines that target human metalloenzymes, with oncology and infectious disease programmes. In oncology it has developed non-hydroxyurea inhibitors of flap endonuclease 1 (FEN1), a DNA-repair metallonuclease with homologous recombination genes among its synthetic lethality partners, and lists QPCTL, which modifies CD47 and CCL2, as a myeloid immunotherapy target. In fiscal year 2026 NCI awarded it USD 399,915 under SBIR award 1R43CA309911-01A1 to combine a selective FEN1 inhibitor with olaparib in PARP inhibitor-resistant ovarian cancer.
