Flare Therapeutics makes small-molecule drugs aimed at transcription factors, the proteins that switch genes on and off. Its two drugs in early trials are for advanced bladder (urothelial) cancer and for prostate cancer that has stopped responding to hormone treatment.
Flare Therapeutics is a privately held Cambridge, Massachusetts company that says it works only on drugging transcription factors, with a chemistry and proteomics engine for finding ligands directly in cells. The company says its pipeline is initially focused on precision oncology. FX-909 is described as a first-in-class oral small-molecule inhibitor of PPARG, the master regulator of the luminal lineage, in a randomised Phase 1b study in PPARG-high locally advanced or metastatic urothelial cancer (NCT05929235, sponsored by Flare, active and not recruiting; one arm adds pembrolizumab). FX-111 is described as a degrader of the transcriptionally active, hormone-bound androgen receptor (AR ON) and is in a Phase 1 study in metastatic castration-resistant prostate cancer (NCT07719361, recruiting). The company also names a partnership with Roche. Third Rock Ventures lists Flare under its Cancer focus and gives 2021 as its year of investment.
