KymaThera is developing K-1728, a pill that blocks the faulty forms of the PI3K-alpha enzyme found in many hormone receptor-positive breast cancers while largely sparing the normal enzyme. It expected to start its first human trial in late 2026.
KymaThera, based in San Diego, develops small-molecule precision therapeutics. Its lead, K-1728, is an oral pan-mutant-selective PI3Kα inhibitor designed to inhibit both kinase-domain and helical-domain PIK3CA mutations while largely sparing wild-type PI3Kα. It is being developed for HR-positive, HER2-negative breast cancer, alone and in combination, and as monotherapy for PI3Kα-driven vascular malformations. In October 2026 the company closed an USD 80 million Series B led by Alta Partners with Venrock, Foresite Capital and J. Wood Capital, bringing total capital raised to more than USD 100 million; it expected to dose the first patient in a phase 1 study in the fourth quarter of 2026. Rob Kania is chief executive.
