Shasqi
Shasqi puts an inert form of a chemotherapy drug into the bloodstream and switches it on only inside the tumour, using a chemical reaction called click chemistry, so the rest of the body sees far less of the drug.
Overview
Shasqi's CAPAC platform (Click Activated Protodrugs Against Cancer) localises a tetrazine-modified biopolymer at the tumour, then infuses a trans-cyclooctene-caged protodrug that is activated by an in vivo click reaction where the two meet. The lead programme SQ3370 delivers a doxorubicin protodrug and completed a first-in-human phase 1 study in advanced solid tumours including soft tissue sarcoma, reporting that the approach allowed doxorubicin exposures well above conventional dosing with manageable toxicity.
