RTOG 9402: PCV chemotherapy before radiotherapy for anaplastic oligodendroglioma, long-term results
Long follow-up of this trial showed that adding PCV chemotherapy to radiotherapy roughly doubled survival, from about seven to fourteen years, in anaplastic oligodendroglioma with loss of chromosomes 1p and 19q, while tumours without the codeletion gained nothing.
Overview
Phase 3 trial of 291 patients with anaplastic oligodendroglioma or oligoastrocytoma randomised to intensive PCV followed by radiotherapy or radiotherapy alone, reported at a median follow-up of 11.3 years.
Overall survival did not differ in the whole cohort, but in 1p/19q-codeleted tumours median survival was 14.7 years with PCV against 7.3 years without (hazard ratio 0.59), while non-codeleted tumours had a median of 2.6 versus 2.7 years.

1p/19q codeletion is a predictive marker: codeleted oligodendroglioma is treated with radiotherapy plus PCV, and the finding helped make the codeletion part of the tumour's definition.
