HCAR3
HCAR3 (Hydroxycarboxylic acid receptor 3) is a gene that drives cell growth when it is altered. The public catalogues list it as an oncogene driver, and it is called a cancer driver by mutation analysis of patient cohorts. Tied to Acute myeloid leukaemia.
Overview
G protein-coupled receptor for 3-hydroxyoctanoate, a fatty acid beta-oxidation intermediate. Signals through the inhibitory G(i)/o family of G proteins. Acts as a negative feedback regulator of adipocyte lipolysis, helping to counterbalance prolipolytic signals during physiological or pathological elevations in beta-oxidation.
IntOGen calls it a driver in 1 cohort (1 activating, 0 loss-of-function), covering Acute Myeloid Leukaemia.

No product in this corpus aims at HCAR3 yet. Drugs fit a pocket that exists only in one shape of the mutant protein and hold it there, off.

Biology
G protein-coupled receptor for 3-hydroxyoctanoate, a fatty acid beta-oxidation intermediate. Signals through the inhibitory G(i)/o family of G proteins. Acts as a negative feedback regulator of adipocyte lipolysis, helping to counterbalance prolipolytic signals during physiological or pathological elevations in beta-oxidation. Acts as a low affinity receptor for nicotinic acid. This pharmacological effect requires nicotinic acid doses that are much higher than those provided by a normal diet. Location: Cell membrane (UniProt). Locus 12q24.31 (HGNC).

Notes
top- Written by scripts/fetch-cancer-genes.ts from CIViC, Open Targets, IntOGen, HGNC and UniProt; the function text is UniProt's, condensed and in UK spelling. Roles: IntOGen calls it an activating (Act) driver in 1 cohort. Evidence tier "cohort-driver" is the strongest of those signals.
- Prevalence not recorded: none of the sources gives a positivity rate.
