TYW2
TYW2 (tRNA wybutosine-synthesising protein 2 homolog) is a gene that drives cell growth when it is altered. The public catalogues list it as an oncogene driver, and it is called a cancer driver by mutation analysis of patient cohorts. Tied to Salivary gland cancers.
Overview
S-adenosyl-L-methionine-dependent transferase that acts as a component of the wybutosine biosynthesis pathway. Wybutosine is a hyper modified guanosine with a tricyclic base found at the 3'-position adjacent to the anticodon of eukaryotic phenylalanine tRNA. Catalyses the transfer of the alpha-amino-alpha-carboxypropyl (acp) group from S-adenosyl-L-methionine to the C-7 position of 4-demethylwyosine (imG-14) to produce wybutosine-86.
IntOGen calls it a driver in 1 cohort (1 activating, 0 loss-of-function), covering Salivary Carcinoma.

- 2 · What goes wrong in cancer
S-adenosyl-L-methionine-dependent transferase that acts as a component of the wybutosine biosynthesis pathway.
- 3 · How drugs use it
No product in this corpus aims at TYW2 yet. Drugs fit a pocket that exists only in one shape of the mutant protein and hold it there, off.

Notes
top- Written by scripts/fetch-cancer-genes.ts from CIViC, Open Targets, IntOGen, HGNC and UniProt; the function text is UniProt's, condensed and in UK spelling. Roles: IntOGen calls it an activating (Act) driver in 1 cohort. Evidence tier "cohort-driver" is the strongest of those signals.
- Prevalence not recorded: none of the sources gives a positivity rate.
