CABONE
CABONE showed that the tablet cabozantinib could shrink or hold back both Ewing sarcoma and osteosarcoma that had come back after standard chemotherapy, the first drug to show meaningful activity in both bone cancers.
Overview
CABONE was a French multicentre phase 2 study of cabozantinib, an inhibitor of MET, VEGF receptors and other kinases, in 90 patients aged 12 or older with advanced Ewing sarcoma or osteosarcoma after standard therapy. The primary endpoint was objective response at six months in the Ewing cohort and objective response or six-month non-progression in the osteosarcoma cohort.
Both cohorts met their predefined activity thresholds, with objective responses in Ewing sarcoma and a high rate of six-month non-progression in osteosarcoma. Toxicity was consistent with cabozantinib in adults, including pneumothorax in some patients with lung metastases. Cabozantinib is now used off-label at relapse and is being tested in combinations in paediatric bone sarcoma trials.
