A European trial testing whether a gentler substitute for high-dose interleukin-2, the drug given after tumour-infiltrating lymphocyte cell therapy, can cut serious side effects while keeping the treatment working, in melanoma, lung and cervical cancer.
PragmaTIL is a Horizon Europe Cancer Mission project (CORDIS 101104684, topic HORIZON-MISS-2022-CANCER-01-03, "Pragmatic clinical trials to optimise treatments for patients with refractory cancers"), coordinated by the Vall d'Hebron Institute of Oncology in Barcelona, running from 1 April 2023 to 31 March 2028 with an EU contribution of about EUR 6.0 million. Tumour-infiltrating lymphocyte adoptive cell therapy normally requires high-dose interleukin-2 to expand and activate the infused cells, and its toxicity is a barrier to use. The project's objectives are to find whether an IL-2 analogue reduces the frequency of grade 3 or higher non-haematological study-related toxicity, to compare quality of life in hospital and over time, and to carry out a health technology assessment and a social return on investment analysis.
ClinicalTrials.gov lists it as NCT06630611, a phase 2 study sponsored by Vall d'Hebron Institute of Oncology in melanoma, non-small-cell lung cancer and cervical cancer, recruiting, with an actual start on 15 January 2025 and a planned enrolment of 40, comparing an IL-2 analogue with high-dose IL-2 after non-myeloablative lymphodepletion and TIL infusion.
