Carrick Therapeutics develops medicines taken by mouth that block enzymes cancer cells use to grow. Its lead drug, samuraciclib, is being tested with hormone therapy in advanced hormone receptor-positive, HER2-negative breast cancer after earlier treatment has stopped working.
Carrick Therapeutics is a clinical-stage oncology company whose lead programme is samuraciclib (CT7001), an oral CDK7 inhibitor discovered at Imperial College London with Cancer Research UK funding. Its main indication is HR-positive, HER2-negative advanced breast cancer after a CDK4/6 inhibitor. In the randomised phase 2 SUMIT-BC trial (NCT05963984, 60 patients) of samuraciclib with fulvestrant against fulvestrant alone, the company reported in December 2025 an overall response rate of 33% versus 14% and median progression-free survival of 7.8 versus 5.6 months in all patients, with larger differences in patients without a TP53 mutation (a pre-specified group), and said it plans a phase 3 trial in 2026. Samuraciclib has also been combined with elacestrant and with vepdegestrant, and the company reports FDA Fast Track designations in CDK4/6 inhibitor-resistant HR-positive breast cancer and in triple-negative breast cancer. Its second programme, CT7439, is an oral CDK12/13 inhibitor that also acts as a molecular glue degrader of cyclin K, in a phase 1/2 trial in solid tumours (NCT06600789). The company has clinical collaborations with Pfizer, Roche, Menarini and Arvinas. It moved its headquarters from Dublin to Boston in June 2024 and keeps teams in Ireland and the UK.
Samuraciclib is an oral cdk inhibitor from Pfizer, in registered phase 2 trials for metastatic cancer, HR-positive / HER2-negative breast cancer.
CT7439 is an experimental pill from Carrick Therapeutics, in phase 1 trials for advanced solid tumours.
