Boundless Bio
Boundless Bio tried to treat cancers driven by extra loops of DNA outside the chromosomes, called ecDNA. Its lead drug did not show enough benefit in early trials, and in 2026 the company agreed to merge with a gene-editing company and leave cancer.
Overview
Boundless Bio is a San Diego (La Jolla) company that developed ecDNA-directed therapies for oncogene-amplified cancers, built on the biology of extrachromosomal DNA. Its programmes included BBI-940, an oral kinesin degrader that entered the phase 1 KOMODO-1 trial in February 2026 for ER-positive/HER2-negative and luminal androgen receptor triple-negative breast cancer, plus CHK1 inhibitors for replication stress and RNR inhibitors intended to prevent ecDNA-mediated resistance to KRAS inhibitors. In June 2026 the company said early clinical data for BBI-940 did not support continuing and announced an all-stock merger with Serapha Bio, a private developer of an in vivo base-editing therapy for alpha-1 antitrypsin deficiency, with a 230 million US dollar private placement co-led by RA Capital and RTW; the combined company is to trade as Serapha Bio (AATD). Boundless raised 100 million US dollars gross in its April 2024 IPO.
