Foghorn Therapeutics makes drugs that act on the machinery that packages DNA and controls which genes are switched on. Its lead drug, partnered with Lilly, targets lung cancers that have lost a gene called SMARCA4.
Overview
Foghorn Therapeutics is a Cambridge, Massachusetts company targeting the chromatin regulatory system through its Gene Traffic Control platform. Its most advanced programme, FHD-909 (LY4050784), is a selective SMARCA2 ATPase inhibitor in phase 1 for SMARCA4-mutant non-small cell lung cancer, partnered with Lilly under a 50/50 US economic split with ex-US royalties; the Lilly collaboration also covers a selective SMARCA2 degrader and other programmes. Wholly owned preclinical programmes include a selective EP300 degrader for multiple myeloma, DLBCL and prostate cancer (IND targeted in 2027), a selective CBP degrader for ER-positive breast cancer, and a selective ARID1B degrader for ARID1A-mutant cancers. Foghorn also signed a collaboration with Merck in July 2020 (15 million US dollars upfront). Foghorn raised 135.2 million US dollars gross in its October 2020 Nasdaq IPO.
