K36 Therapeutics
K36 Therapeutics makes pills that block an enzyme called NSD2 (also known as MMSET), which is overactive in about one in five multiple myeloma cases because of a chromosome swap. It is also testing the approach in prostate cancer.
Overview
K36 Therapeutics is a Cambridge, Massachusetts clinical-stage company and the first to target the histone methyltransferase NSD2 (MMSET/WHSC1). Gintemetostat (KTX-1001) is a first-in-class, oral, selective catalytic NSD2 inhibitor in the K36-MMSET-001 trial in relapsed or refractory multiple myeloma with the t(4;14) translocation; the company reports target engagement, an encouraging safety profile and clinical activity in heavily pretreated patients, with expansion cohorts combining it with carfilzomib, mezigdomide or pomalidomide. KTX-2001, a second oral NSD2 inhibitor, is in the K36-STRIKE-001 trial in metastatic castration-resistant prostate cancer alone and with darolutamide. K36 launched in December 2021 with a 30 million US dollar Series A co-led by F-Prime Capital and Atlas Venture, and raised a 70 million US dollar Series B in June 2023 led by Nextech Invest with Bristol Myers Squibb participating.
