ORIC Pharmaceuticals develops pills designed to beat the ways cancers resist treatment. One blocks a gene-silencing complex in prostate cancer; the other targets mutated EGFR and HER2 in lung cancer and can reach the brain.
Overview
ORIC Pharmaceuticals is a South San Francisco clinical-stage oncology company focused on mechanisms of therapeutic resistance. Rinzimetostat (ORIC-944) is a potent, selective allosteric inhibitor of PRC2 acting through the EED subunit rather than EZH2, designed for better drug properties and to avoid EZH1 bypass or EZH2 mutation; it is in development for prostate cancer in combination with androgen receptor pathway inhibitors. Enozertinib (ORIC-114) is a brain-penetrant inhibitor of EGFR exon 20 insertion and other atypical EGFR and HER2 alterations in non-small cell lung cancer, including a clinical collaboration with Johnson & Johnson combining it with subcutaneous amivantamab in first-line EGFR exon 20 NSCLC. ORIC completed its Nasdaq IPO in April 2020 with gross proceeds of 138 million US dollars.
