Synnovation Therapeutics develops targeted pills for genetically defined cancers. Its PARP1 blocker SNV1521, built to reach the brain, is in early trials including one with a RAS drug in pancreatic cancer; in 2026 Novartis agreed to buy its breast cancer PI3K-alpha programme for USD 2 billion upfront.
Synnovation Therapeutics, with its main office in Wilmington, Delaware, launched on 24 January 2024 with a USD 102 million Series A led by Third Rock Ventures with Nextech, Lilly Asia Ventures, Sirona Capital and Cormorant Asset Management, to advance small-molecule precision oncology medicines. SNV1521 is a PARP1-selective, brain-penetrant inhibitor; its first patient was dosed on 11 March 2024, phase 1 data were presented at ESMO 2025, and a clinical collaboration with DualityBio on the HER3 antibody-drug conjugate DB-1310 was announced in October 2025. A further phase 1 study combines SNV1521 with the RAS(ON) inhibitor daraxonrasib in advanced pancreatic cancer. SNV4818, a pan-mutant-selective PI3Kα inhibitor, dosed its first patient on 24 February 2025; on 20 March 2026 Novartis agreed to acquire Pikavation Therapeutics, the wholly owned subsidiary holding the PI3Kα programmes, for USD 2 billion upfront and up to USD 1 billion in milestones, with Synnovation keeping SNV1521 and its other subsidiaries.
