SIL204 is a gene-silencing medicine from Silexion Therapeutics that stops cancer cells making the KRAS protein that drives them. It is given with chemotherapy and is in a larger trial in pancreatic cancer.
SIL204 is Silexion Therapeutics' second-generation siRNA against KRAS. It is given directly into the tumour and systemically by subcutaneous injection, together with standard chemotherapy. Silexion received Israeli and German approvals in the second quarter of 2026 for a phase 2/3 trial in locally advanced pancreatic cancer and initiated it in Tel Aviv in late July 2026. A first-generation siRNA from the company was tested with chemotherapy in an earlier phase 2 trial.
A second-generation small interfering RNA engineered to suppress expression of the KRAS oncogene, preventing production of the oncogenic protein, as described by Silexion Therapeutics. Connects to KRAS.
