A chemistry that enriches the tumour's mutated DNA before sequencing found lung cancer DNA at a few parts per million with far less sequencing than usual.
Enspyre is a ctDNA enrichment method. Tumour and, where available, matched normal tissue from eight lung cancer patients were whole-genome sequenced; custom capture panels ran on 72 samples of patient plasma diluted into healthy donor plasma (nine per subject) and eight healthy donor controls. Enspyre reached 100 per cent (6 of 6) detection down to 10 parts per million and called residual disease as low as 5 parts per million with no false positives, at a median DNA input of 7.67 ng, without molecular barcodes, with a median of 9.6 million read pairs per sample. The authors note that larger studies are needed.
Enrichment may make parts-per-million testing possible on small sequencers, though only eight patients were studied.
