OriMIRACLE S identifies tumour-specific variants by whole-exome sequencing of tumour and germline DNA, selects clonal SNVs with the OriSelector algorithm for patient-specific multiplex PCR panels, and calls a sample positive when three sites are detected, giving nearly 100 per cent sample-level sensitivity and specificity with panels of 21 to 30 variants. In 127 patients with gastrointestinal tumours, residual disease was positive before surgery in 18 of 26 and after surgery in 24 of 82; positivity was 33.33 per cent in gastric adenocarcinoma and 32.26 per cent in colorectal cancer.
A residual disease test built on clonal variants only, which detects the trunk of the tumour and by design not its branches.
