The clinician half of the partnership that turned a liposome chemistry idea into Doxil, and the person who did the trials that showed it behaved differently in patients from free doxorubicin.
Alberto Gabizon worked with Yechezkel Barenholz at the Hebrew University on pegylated liposomal doxorubicin and then took it into patients, establishing the pharmacokinetics that distinguish it from free doxorubicin: a long circulation half-life, accumulation in tumour tissue, and a toxicity profile in which hand-foot syndrome and mucositis replace alopecia and cardiac damage. The product was approved by the FDA in 1995 and remains standard in relapsed ovarian cancer, used as a single agent or with carboplatin, trabectedin or bevacizumab, and as the comparator arm in modern trials including MIRASOL. He continues to work at Shaare Zedek Medical Center on nanomedicine pharmacology and combination strategies.
