EGFRvIII

EGFRvIII is a mutant, tumour-only version of the EGFR receptor found in about a third of glioblastomas. It is an ideal-looking target, yet every drug against it has failed so far.

In-frame deletion of exons 2-7 creating a constitutively active receptor with a tumour-specific junctional epitope. Targeted by rindopepimut (ACT IV negative), EGFRvIII CAR-T (antigen loss), bispecifics (AMG 596), and ADCs (depatuxizumab mafodotin, INTELLANCE-1 negative). Expression is heterogeneous and frequently lost at recurrence.
