APTN-101 is a two-armed antibody from Adaptin Bio that steers a patient's own T cells against brain tumour cells carrying a faulty growth receptor. It is in a first trial in people with glioblastoma at Duke University.
APTN-101 is Adaptin Bio's EGFRvIII x CD3 bispecific (BRiTE). The FDA accepted an investigator-initiated IND for a single-dose trial in May 2023 and a multi-dose amendment in October 2025; institutional review board approval for the multi-dose trial in glioblastoma came in March 2026, and Duke University opened the investigator-initiated phase 1 trial, "A Phase I Study of hEGFRvIII-CD3 bi-scFv (BRiTE) in Patients with WHO Grade IV Malignant Glioma", in August 2026.
A fully human bispecific single-chain antibody that binds EGFRvIII, a mutant form of EGFR found on tumour cells, and CD3 on T cells, redirecting the patient's T cells to kill the tumour; Adaptin Bio's BRiTE approach uses activated T cells to carry it into the brain. Connects to CD3.
