RTOG 9601
RTOG 9601 showed that adding two years of hormone therapy to salvage radiotherapy after prostate surgery lengthens survival, mainly in men with higher PSA levels at relapse.
Overview
RTOG 9601 randomised 760 men with a rising PSA after radical prostatectomy to salvage radiotherapy (64.8 Gy) with 24 months of bicalutamide 150 mg or placebo. Twelve-year overall survival was 76.3 percent with bicalutamide and 71.3 percent with placebo (hazard ratio 0.77), with fewer prostate cancer deaths and metastases; the benefit was concentrated in men with PSA above 0.7 ng/mL, and gynaecomastia was common (Shipley and colleagues, New England Journal of Medicine 2017).
- 76.3 vs 71.3 out of 100 alive at 12 years with Salvage radiotherapy plus bicalutamide compared with Salvage radiotherapy plus placebo; 5 more per 100.
- Roughly one extra person helped for every 20 treated. That is a rough figure taken from the two percentages, not a guarantee for any one person.
- Overall survival counts deaths from any cause, so it is the most direct measure of whether a treatment helps people live longer.
- These results apply to the people the trial enrolled: Rising PSA after prostatectomy: salvage radiotherapy with two years of bicalutamide versus salvage radiotherapy with placebo. People in a different situation may not see the same effect.
