ZEN-3694 with abemaciclib in NUT carcinoma, breast cancer and other solid tumours (NCI phase 1)
A chemotherapy-free combination for NUT carcinoma: a BET inhibitor with a CDK4/6 inhibitor, on the reasoning that both act on the MYC-driven growth programme the fusion switches on.
Overview
BRD4-NUT keeps MYC and other growth genes switched on; BET inhibition alone gives short responses. This NCI phase 1 study adds abemaciclib, a CDK4/6 inhibitor, to ZEN003694 in patients with NUT carcinoma, advanced breast cancer and other solid tumours, to find a tolerable dose and look for activity. It is recruiting.
