Rgenta Therapeutics makes tablet medicines that act on a gene's RNA message rather than its protein; its lead drug switches off MYB, the driver of adenoid cystic carcinoma, a rare cancer of the salivary glands.
Rgenta Therapeutics, Inc., of Woburn, Massachusetts, develops oral small-molecule RNA-targeting medicines: small molecule glues that stabilise the interaction between a target RNA and the splicing machinery so that the target RNA is degraded. Its lead programme RGT-61159 inhibits production of the oncogenic transcription factor MYB and is in a phase 1a/b trial in relapsed or refractory adenoid cystic carcinoma and colorectal cancer (NCT06462183); it received FDA orphan drug designation for adenoid cystic carcinoma in September 2025, and in October 2026 the company announced an end-of-phase 2 meeting with the FDA and alignment on a phase 3 design. Preclinical work extends RGT-61159 to acute myeloid leukaemia, and the company has a CNS programme and an RSwitch gene-regulation technology. In January 2024 it joined the Leukemia & Lymphoma Society Therapy Acceleration Program, which provided strategic funding. Its first registered cancer trial, NCT06462183, was posted on ClinicalTrials.gov in June 2024.
