Rondo Therapeutics makes two-armed antibodies that grab a tumour protein with one arm and give T cells an extra activation signal with the other, so the immune system attacks solid tumours. Its first drug, RNDO-564, is in an early trial for bladder cancer and other tumours that carry Nectin-4.
Rondo Therapeutics, at 25801 Industrial Boulevard, Hayward, California, was co-founded by chief executive Shelley Force Aldred and chief scientific officer Nathan Trinklein and develops immune-cell engaging bispecific antibodies for solid tumours using an NGS-based antibody discovery platform with a fixed light-chain format. The lead candidate RNDO-564 is a CD28 x Nectin-4 bispecific, tested alone and with pembrolizumab in a phase 1/1b study in bladder cancer and other Nectin-4 associated solid tumours, including non-small cell lung, cervical, head and neck and triple-negative breast cancers. Earlier programmes pair CD3 and CD28 arms with undisclosed targets for ovarian, endometrial and renal cancers, and the company announced a collaboration with Lilly on CD28 bispecific antibodies in December 2024. Its first registered cancer trial, NCT07218003, was posted on ClinicalTrials.gov in October 2025. In March 2022 it closed a USD 67 million Series A co-led by Red Tree Venture Capital and Canaan Partners with Johnson & Johnson Innovation (JJDC), Novo Holdings and SV Health Investors.
