NST-628 is an oral small-molecule molecular glue from Nested Therapeutics. A non-degrading, fully brain-penetrant pan-RAF/MEK molecular glue that holds RAF and MEK (MEK1/2) together, hitting several parts of the RAS/MAPK pathway at once. Its furthest stage is phase 1, for MAPK pathway-mutated or -dependent advanced solid tumours, melanoma, non-small cell lung cancer and glioma. ClinicalTrials.gov registers it in 2 studies linked below. Phase 1 NCT06326411 recruiting; an investigator-sponsored early phase 1 in recurrent grade 4 glioma (NCT07784985, Barrow Neurological Institute, Nested as collaborator) is not yet recruiting. Whether it works in people has not been established unless a trial below reports it.
