Exact Sciences' tissue test for advanced solid tumours that reads every gene's DNA and RNA and compares the tumour with the patient's normal DNA.
OncoExTra is Exact Sciences' comprehensive genomic profiling test for advanced, metastatic, refractory, relapsed or recurrent solid tumours. It sequences the whole exome of the tumour against a matched normal sample and the whole transcriptome of the tumour, and reports mutations and fusions associated with FDA-approved therapies, tumour mutational burden and microsatellite status, and clinical trial options. Exact Sciences cites the 2021 validation of GEM ExTra as the test's own validation: that assay detects single-nucleotide variants, insertions and deletions, focal copy number changes and the TERT promoter region, and its RNA sequencing finds fusions and transcripts such as AR-V7, EGFR vIII and MET exon 14 skipping. In a retrospective analysis of 1,509 reports that Exact Sciences quotes, 83.9% showed at least one clinically actionable variant. The test was developed by Genomic Health, an Exact Sciences subsidiary, is performed at its Phoenix laboratory, and has not been cleared or approved by the FDA.
Whole-exome DNA sequencing of the tumour and of a matched normal sample, so inherited variants can be subtracted, plus whole-transcriptome RNA sequencing of the tumour for gene fusions and selected abnormal transcripts.
