Toceranib (Palladia) is a tablet for dogs, not people, approved by the US Food and Drug Administration in 2009 to treat mast cell tumours of the skin that have come back. It blocks the same family of growth signals as the human cancer drug sunitinib.
Palladia (toceranib phosphate) tablets were approved by the FDA Center for Veterinary Medicine on 22 May 2009 under NADA 141-295, for the treatment of Patnaik grade II or III, recurrent, cutaneous mast cell tumors with or without regional lymph node involvement in dogs; the sponsor was Pharmacia & Upjohn Company, a division of Pfizer, and the current label names Zoetis as distributor.
The pivotal study was a multicentre, placebo-controlled, double-blind trial in dogs with recurrent mast cell tumours after surgery (London et al., Clin Cancer Res 2009). In the six-week blinded phase the objective response rate was 37.2% with toceranib (86 dogs) against 7.9% with placebo (63 dogs). The label describes toceranib as an inhibitor of VEGFR2, PDGFR and KIT (the trial report names PDGFRB), the receptors that the human drug sunitinib also blocks; it was developed under the code SU11654. It is a veterinary medicine and has no human approval.
An oral inhibitor of split-kinase receptor tyrosine kinases; the label states it inhibits Flk-1/KDR (VEGFR2), PDGFR and the stem cell factor receptor KIT, and canine mast cell tumours are frequently driven by activating KIT mutations. Connects to KIT, VEGF / VEGFR and PDGFRB.
1.Many mast cell tumours in dogs grow because the KIT receptor is switched on by a mutation
