Boston sarcoma doctor who first showed that mTOR inhibitors shrink perivascular epithelioid cell tumours and then led AMPECT, the trial that made nab-sirolimus the first approved treatment for these rare cancers.
Overview
Andrew Wagner is a medical oncologist in the sarcoma centre at Dana-Farber Cancer Institute. He was first author of the 2010 Journal of Clinical Oncology report showing responses to sirolimus in malignant perivascular epithelioid cell tumours, linking their TSC1/TSC2 loss to mTOR dependence, and of the 2021 Journal of Clinical Oncology report of AMPECT, the phase 2 trial in which nab-sirolimus produced durable responses in advanced malignant PEComa, the basis of the first approval for the disease.
Nab-sirolimus is the approved first-line treatment for advanced malignant PEComa, and TSC1/TSC2 testing helps predict response.
mTOR inhibition became the therapeutic strategy for PEComa, culminating in the AMPECT trial and approval of nab-sirolimus.
