Tissue factor
Tissue factor is a clotting protein that cancers abnormally display on their surface, used as an ADC target in cervical cancer.
Tissue factor is overexpressed in cervical, pancreatic, and other solid tumours. Tisotumab vedotin (Tivdak) is approved in recurrent cervical cancer (innovaTV 301). Topoisomerase-I payload successors (XNW28012) are in phase 3.
- Target · the protein and the cell it sits on
- Drug · antibody, small molecule, cell or radioligand
- Effect · signal, damage or kill

- 1 · What it is

- 2 · What goes wrong in cancer
Initiates the extrinsic coagulation cascade; tumour expression drives thrombosis and angiogenesis.
- 3 · How drugs use it
1 product aims at Tissue factor: antibody-drug conjugates. Because it sits on the outside of the cell, it can be reached from the bloodstream: antibodies flag the cell, ADCs deliver a toxin, radioligands deliver radiation, and CAR-T or bispecifics bring a T cell.

| Cervical cancer | 90-95% | IHC, any expression | innovaTV did not select on TF | Wikipedia |
| Pancreatic ductal adenocarcinoma | 50-80% | IHC, any expression | Wikipedia |
