Repertoire Immune Medicines designs off-the-shelf medicines that steer the body's T cells to a marker found on cancer cells. Its lead, RPTR-1-201, is in an early trial in advanced solid tumours and has FDA Fast Track status for triple-negative breast cancer.
Repertoire Immune Medicines was founded in 2019 by Flagship Pioneering and operates in Cambridge, Massachusetts, and Zurich. Its DECODE platform maps T cell receptor (TCR) and epitope pairs from tumour-infiltrating T cells to design TCR bispecifics (an engineered TCR joined to an anti-CD3 domain that redirects T cells to tumour cells) and off-the-shelf mRNA cancer vaccines; it also develops tolerising therapies for autoimmune disease. Its lead, RPTR-1-201, is in a phase 1/2 trial alone and with an anti-PD-1 antibody in advanced solid tumours (NCT07293754), with cohorts listed for microsatellite-stable colorectal cancer, non-small-cell lung cancer, head and neck squamous cell carcinoma, triple-negative breast, pancreatic, gastric, bladder, cervical and oesophageal cancer. In May 2026 the FDA granted Fast Track designation to RPTR-1-201 for HLA-A*02:01-positive adults with unresectable or metastatic triple-negative breast cancer after standard therapies. Earlier programmes in development include RPTR-1-301 (HPV-positive cancers), RPTR-1-004 (prostate cancer) and half-life-extended RPTR-1-005 and RPTR-1-006. The company has partnerships with Bristol Myers Squibb, Genentech, Pfizer and Eli Lilly.
RPTR-1-201 is an experimental bispecific protein from Repertoire Immune Medicines, in phase 1 trials for advanced solid tumours in people with the HLA-A*02:01 tissue type.
