Rock425 is an experimental immunotoxin from Rock Immune that carries a disabled diphtheria toxin to malignant T cells. It is being developed for cutaneous T-cell lymphoma and has not yet been tested in people.
Rock425 (CCR4-IL2 IT) is Rock Immune's lead candidate: a genetically engineered protein containing an anti-human CCR4 scFv and human IL-2 fused to truncated diphtheria toxin, expressed in the company's diphtheria toxin-resistant Pichia pastoris system. Rock Immune reports preclinical efficacy in a mouse model of human cutaneous T-cell lymphoma. No registered trial was found.
An anti-CCR4 single-chain antibody fragment and human IL-2 fused to a truncated diphtheria toxin, so that the toxin is carried into cells bearing CCR4 or the IL-2 receptor, as described by Rock Immune. Connects to CCR4.
1.Rock425's antibody arm binds CCR4 on the surface of the cancer cell.
