A small-molecule inhibitor of the ATPase of DNA polymerase theta (Polθ), a synthetic-lethal target linked to BRCA mutations and other genomic changes; tested alone and with olaparib. Its furthest stage is phase 1, for epithelial ovarian cancer, metastatic breast cancer, metastatic castration-resistant prostate cancer and pancreatic adenocarcinoma. ClinicalTrials.gov registers it in one study linked below. Phase 1 POLAR NCT06560632 terminated ('Sponsor decided to terminate study early'); Repare said in November 2025 it would no longer report POLAR topline data and was seeking to license the programme. Whether it works in people has not been established unless a trial below reports it.
