A large European trial asking whether men with prostate cancer that has spread, who respond well to modern hormone treatment, can take planned breaks from it without shortening their lives, with fewer side effects and lower cost.
DE-ESCALATE is a Horizon Europe Cancer Mission project (CORDIS 101104574, topic HORIZON-MISS-2022-CANCER-01-03), coordinated by the EORTC in Brussels, running from 1 June 2023 to 31 May 2028 with an EU contribution of about EUR 7.2 million. Its CORDIS objective is to evaluate whether intermittent intensified androgen deprivation therapy in metastatic prostate cancer is not inferior to continuous treatment in oncological benefit while reducing side effects and resource use and improving quality of life, with a design meant to detect early any negative effect on overall survival; the consortium includes oncologists, surgeons, health economists and patient representatives.
ClinicalTrials.gov lists it as NCT05974774, "Intermittent Androgen Deprivation Therapy in the Era of AR Pathway Inhibitors", a phase 3 study sponsored by the EORTC in prostate cancer, recruiting, with an actual start on 23 May 2025 and a planned enrolment of 1,600.
