TAPPAS
TAPPAS tested whether adding an antibody against endoglin, a protein on the abnormal blood vessel cells that make up angiosarcoma, would improve on the standard tablet pazopanib; it did not, and the trial was stopped for futility.
Overview
Angiosarcoma cells express endoglin (CD105), and TRC105 (carotuximab) is an antibody against it that had shown responses in combination with pazopanib in an earlier study. TAPPAS was an adaptive phase 3 trial randomising 128 patients with advanced angiosarcoma, cutaneous and non-cutaneous, to pazopanib with or without TRC105, with progression-free survival as the primary endpoint.
An interim analysis found no improvement in progression-free survival with the combination and the trial was stopped for futility. It remains one of the few randomised trials ever completed in angiosarcoma, and its control arm data on pazopanib are a reference for the disease.
