Triana Biomedicines
Triana Biomedicines designs molecular glues that make cancer cells destroy their own faulty proteins. Its first drug, TRI-611, removes the ALK fusion protein that drives a subset of lung cancers and has entered human trials.
Overview
Triana Biomedicines is a Lexington, Massachusetts company with a target-first molecular glue discovery platform that pairs disease targets with the most suitable of more than 600 E3 ubiquitin ligases. Its lead candidate, TRI-611, is an oral, brain-penetrant molecular glue degrader that brings ALK fusion proteins to cereblon independently of the kinase active site, intended to overcome resistance, CNS metastases and tolerability limits of ALK inhibitors in ALK-positive non-small cell lung cancer; it has entered the clinic. A CCNE1 (cyclin E1) degrader programme targets CCNE1-amplified tumours and a subset of HR-positive/HER2-negative breast cancers. Triana launched in April 2022 with 110 million US dollars in total funding (seeded by RA Capital and Atlas Venture, Series A co-led by Lightspeed) and closed a 120 million US dollar Series B in December 2025 co-led by Ascenta Capital and Bessemer Venture Partners.
